루퍼스는 베커, 밀러 등과 함께 연희전문학교 수물과를 창설하여 우리나라의 초기 근대 자연과학 교육 체계를 세우는데 기여한 선교사였다. 루퍼스는 1907년 한국에 와서 숭실학당에서 수학 및 과학 교사로 활약하면서 한국의 천문학사를 연구하였다. 그 첫 결과로 1913년에 조선 태조의 석각천문도인 천상열차분야지도를 분석한 논문을 발표하였고, 1915년에 이 내용을 정리하여, 미국 천문학 전문지에 게재하여 한국 천문학을 처음으로 세계에 알렸다.
1912년 이후에는 서울에서 기독교연합대학(Chosen Christian College, 후에 연희전문학교) 설립에 헌신하였고, 1917년까지 연희전문학교 교수로 봉직하였다. 1917년 귀국 후에는 미시간대학교 천문학과 교수와 천문대장을 맡았다. 1935년에 안식년을 얻어 1년간 우리나라에 다시 와서 연희전문에 머무르면서 한국천문학에 대한 책을 출간하였다.
루퍼스는 연희전문, 연세대학교의 천문학은 물론이고 한국 천문학의 정착에 기여한 대표적인 선교사 과학자였다. 특히 그는 미시간대학에 재직하던 1922년에 연희전문학교 수물과 1회 졸업생 이원철을 대학원생으로 받아들여 박사학위 논문을 지도하였고, 이원철은 한국의 최초 이학 박사가 되어 이후 연세의 천문학 연구를 이어갔다.
